Can I play Fallout 3 with a PCI graphics card or will I need a PCI-E graphics card?

If you’re computer doesn’t have an AGP or a PCI-Express slot on the motherboard then it’s probably far too old to support Fallout 3 anyway.
Check out “Can You Run it”: http://cyri.systemrequirementslab.com/CYRI/ and see how the rest of your PC fares.
